Zalux is a stimulant drug marketed as a 'wakefulness promoting agent' and is one of the stimulants used in the treatment of narcolepsy. Narcolepsy is caused by dysfunction of a family of wakefulness-promoting and sleep-suppressing peptides, the orexins, whose neurons are activated by modafinil. The prexin neuron activation is associated with psychoactivation and euphoria. The exact mechanism of action is unclear, although in vitro studies have shown it to inhibit the reuptake of dopamine by binding to the dopamine reuptake pump, and lead to an increase in extracellular dopamine. Zalux activates glutamatergic circuits while inhibiting GABA.

Also, it has been used in the treatment of hypersomnia, a disorder in which patients lack the capacity for meaningful sleep and may require ten or more hours per day. Recent studies have have found that modafinil may help recovering cocaine addicts fight their addiction.

Uses

Zalux is a stimulant used to improve wakefulness in patients with sleep apnea, narcolepsy, or shift work disorder.

To improve wakefulness in patients with excessive daytime sleepiness (EDS) associated with narcolepsy.

Zalux is also used to associated treatment for these conditions: Attention Deficit Hyperactivity Disorder (ADHD), Fatigue, Major Depressive Disorder (MDD), Narcolepsy, Obstructive Sleep Apnea (OSA), Shift-work related sleep disturbance

How Zalux works

The exact mechanism of action is unclear, although in vitro studies have shown it to inhibit the reuptake of dopamine by binding to the dopamine reuptake pump, and lead to an increase in extracellular dopamine. Zalux activates glutamatergic circuits while inhibiting GABA. Zalux is thought to have less potential for abuse than other stimulants due to the absence of any significant euphoric or pleasurable effects. It is possible that modafinil acts by a synergistic combination of mechanisms including direct inhibition of dopamine reuptake, indirect inhibition of noradrenalin reuptake in the VLPO and orexin activation. Zalux has partial alpha 1B-adrenergic agonist effects by directly stimulating the receptors.

Food Interaction

  • Take with or without food. The absorption is unaffected by food.

[Minor] Administration with food may delay the absorption of modafinil (the racemate) and armodafinil (the R-enantiomer) without significantly affecting their overall bioavailability.

According to the product labeling, modafinil's absorption may be delayed by approximately one hour if taken with food.

Similarly, the time to reach peak plasma concentration (Tmax) of armodafinil may be delayed by approximately 2 to 4 hours in the fed state.

Zalux Hypertension interaction

[Major] CNS stimulant medications have shown to increase blood pressure, and their use might be contraindicated in patients with severe hypertension.

Caution should be used when administering to patients with preexisting high blood pressure and other cardiovascular conditions.

All patients under treatment should be regularly monitored for changes in blood pressure and heart rate.
